Optional Depletion of High-Abundance Proteins with Agilent Multiple Affinity Removal Spin Cartridges
For human samples, the spin cartridge removes albumin, IgG, anti-trypsin,
IgA, transferrin and haptoglobin with a single device. For mouse/rat samples,
it removes albumin, lgG, and transferrin with a single device. When crude
biological samples are passed through the cartridge, the targeted
high-abundance proteins are simultaneously removed by the immobilized
antibodies. Selective immunodepletion provides an enriched pool of
low-abundance proteins for downstream proteomics analysis.
For human samples, specific removal of six high-abundance proteins depletes
approximately 85% to 90% of the total protein mass from serum. For mouse/rat
samples, removal of three high-abundance proteins depletes approximately
80% of the total protein mass from serum. The low-abundance proteins in the
flow-through fractions can then be studied more readily. Removal of
high-abundance proteins enables improved resolution and dynamic range for
one-dimensional gel electrophoresis (1DGE), two-dimensional gel
electrophoresis (2DGE) and liquid chromatography/mass spectrometry
(LC/MS). Depending upon the downstream applications, the collected
flow-through fractions may require concentration.
